چکیده

The effects of induction and of transient and catabolite repression on lac-specific RNA in cells of Escherichia coli were investigated. The rate of Zac-specific RNA synthesis increases linearly for 3 min after induction and then becomes constant. The accumulation of RNA complementary to the promoter proximal tat 2 gene begins before accumulation of RNA complementary to the promoter distal lac Y-A genes. When transient repression is imposed upon the cells by the addition of glucose, the lac Z-specific RNA begins to decay before the lac Y-A-specific RNA. Catabolite repression reduces the rate of formation of lac-specific RNA relative to the rate of formation of bulk RNA. Both lac Z-specific RNA and lac Y-A-specific RNA are equally aBected. These results confirm the concept that the sole effect of transient repression and of catabolite repression is the inhibition of the initiation-of RNA synthesis at the lac promoter site.

Regulatory Gene Control of Transcription of the L-Arabinose Operon in Escherichia coZi*

The E-arabinose operon is under both positive and negative control by the product of its regulatory gene, UrQc. We have measured transcription of the arabinose genes by hybridization of 3H-labeled messenger RNA to single-stranded DNA from an arabinose transducing phage in order to study the effect of the regulatory gene on transcription. In strains in which the arabinose regulatory gene is comp...

Role of cyclic adenosine 3',5'-monophosphate and the cyclic adenosine 3',5'-monophosphate receptor protein in the initiation of lac transcription.

Optimal transcription of the lac operon in a defined purified in vitro system is dependent upon the concentrations of cyclic adenosine 3’,5’-monophosphate (cyclic AMP), the cyclic AMP-binding protein (CRP) and RNA polymerase. The concentration of CRP required for maximal Zac transcription varies proportionately with the concentration of lac DNA used as template. Maximal Zac transcription is obt...

Interactions of Tryptophan Synthetase Subunits in Escherichia coZi Containing Mutationally Altered p2 Subunits*

The (Y and /I2 subunits of tryptophan synthetase are specified by the A and B genes, respectively, of the trp operon in Escherichia coli. Missense mutations in trpB result in catalytically inactive /I2 subunits. In some cases, these mutations also diminish the catalytic and /I2 subunit stimulating activities of the (Y subunit in cell extracts. Analysis of repressor-negative (trpR) strains havin...

Molecular Study of Phase Variation of Type 1 Fimbriae in Uropathogenic Escherichia coli O44 Serotypes during Touching with Solid Surfaces

Background & Aims: Type 1 fimbriae is the most common adhesion factor in urine tract infection. In this Study, presence of virulence genes in isolated strains of uropathogenic E.Coli, O serotyping and molecular detection of phase variation of type 1 fimbriae were assessed during solid surfaces exposure. Methods: Isolated E.coli from urine samples of patients were serotyped by using serologic me...
